Abstract Sperm cryopreservation has become an indispensable tool in reproductive biology. However, frozen/thawed semen has a short lifespan due to loss of sperm cell integrity. To better understand which sperm cell structures are compromised by the cryopreservation process and apoptosis markers, the sperm of five healthy mature dogs was analyzed in this study. Analysis was performed after collection, cooling, and thawing via computer assisted sperm analyzer (CASA) and evaluation of membrane fluidity and permeability, phosphatidylserine translocation (Annexin V), membrane integrity, mitochondrial membrane potential, membrane lipid peroxidation (LPO) and activity of the apoptotic markers caspases 3 and 7 by flow cytometry. Cryopreservation decreased total and progressive motility and the percentage of rapid sperm (P < 0.01). Damage to sperm cells was confirmed by Annexin V (P <0.01), indicating that capacitation-like changes were induced by the cryopreservation procedures. An increase in sperm membrane fluidity was also noted in frozen/thawed samples (P < 0.01). Plasma and acrosomal cell membranes were affected (P < 0.01), with decreases in the subpopulation displaying high membrane potential (P < 0.01). Membrane LPO was increased in thawed sperm compared to cooled sperm (P < 0.05) but was not different from that in fresh sperm. No differences were observed in caspase 3 and 7 activity after cooling, freezing, or thawing. In conclusion, total and progressive motility, plasma membrane integrity and mitochondrial membrane potential suffered from the deleterious effects caused by cryopreservation, unlike the activity of caspases that remained stable during the freezing process.

Resumo Diferentemente de outras espécies domésticas, a maturação in vitro (MIV) de oócitos caninos apresenta sucesso limitado. O objetivo deste estudo foi avaliar o efeito do HEPES na maturação in vitro de oócitos caninos obtidos de cadelas em diestro e anestro. Os ovários foram coletados, isolados assepticamente e transportados refrigerados a uma temperatura de 4 ºC. Os complexos cumulus-oócito (CCOs), provenientes das duas fases do ciclo estral, foram submetidos a dois tratamentos: meio TCM-199 com adição de 25 mM de HEPES (GT) e meio sem suplementação (GC). Depois de 72 horas de maturação, os CCOs foram desnudados, fixados e corados com HOESCHT 33342 para avaliação da maturação nuclear. Os oócitos obtidos da fase de anestro e diestro do GT demonstraram, em relação ao grupo GC, maior frequência de oócitos nos estágios de M-II (p<0,01). Comparando-se os diferentes status reprodutivos, observou-se que os oócitos obtidos da fase de diestro apresentaram índices maiores de QVG e M-II. Nossos resultados demonstraram que o HEPES preserva a viabilidade e morfologia oocitária, indispensáveis para a aquisição da competência meiótica, potencializando as taxas de M-II, e que os oócitos obtidos da fase de diestro estão mais aptos a completarem a maturação oocitária.

Abstract Unlike other domestic animals, in vitro maturation (IVM) of canine oocytes still has limited success. The aim of this study was to evaluate the effect of HEPES on the in vitro maturation of dog oocytes in anestrus and diestrus. Ovaries were collected, aseptically isolated and transported refrigerated at a temperature of 4ºC. Cumulus-oocyte complexe (COCs) from the two phases of estrous cycle were subjected to two treatments: medium TCM-199 with addition of 25 mM HEPES (TG) and medium without supplementation (CG). After 72 h of maturation, COCs were denuded, fixed and stained with Hoechst 33342 to assess nuclear maturation. The oocytes obtained from the anestrus and diestrus phases of the TG showed a higher frequency of oocytes in metaphase II (M-II) (p <0.01) stage.Comparing the different reproductive status, it was observed that the oocytes obtained from the diestrus phase presented higher rates of GVBD (germinal vesicle breakdown) and M-II. Our results showed that HEPES preserves the viability and oocyte morphology essential for the acquisition of meiotic competence, increasing the M-II rates and that the oocytes obtained from the diestrus phase are better able to complete oocyte maturation.

O objetivo do presente estudo foi determinar a ação de diferentes concentrações de toxina botulínica tipo A (TB-A) sobre os parâmetros seminais, perfis bioquímicos e proteicos do plasma seminal de cães com hiperplasia prostática benigna (HPB). Dezoito cães hígidos, não orquiectomizados com HPB foram divididos em três grupos, os quais foram submetidos à injeção intra-prostática de solução salina (grupo controle - GC), 250UI (GI) ou 500UI (GII) de TB-A. Amostras seminais foram coletadas previamente aos tratamentos e após 2, 4 e 8 semanas. Os parâmetros seminais assim como os valores de pH e concentrações de proteínas totais (TP), cloretos totais (CT), cálcio (Ca), potássio (K), sódio (Na) do plasma seminal foram mensurados após as coletas. O perfil proteico do fluido prostático foi estabelecido por meio de eletroforese SDS-PAGE. Não foram constatadas diferenças significativas quanto aos parâmetros espermáticos e perfil bioquímico do plasma seminal intragrupos e intergrupos (P>0,05). À SDS-PAGE foram identificadas 31 bandas proteicas com pesos moleculares de 3,9 a 106,2kDA, em todos os tratamentos e durante todo o período de avaliação. Dessa forma, concluiu-se que, independentemente da dose utilizada, a injeção intra-prostática de TB-A não altera os parâmetros seminais, assim como os perfis bioquímico e proteico do plasma seminal de cães com HPB.

This study aimed to determine the effects of different concentrations of botulinum toxin type A (BT-A) on semen parameters, and seminal plasma biochemical and protein profiles of dogs with benign prostatic hyperplasia (BPH). Eighteen sexually intact male dogs with BPH were randomly divided in three groups, and received an intraprostatic injection of saline solution (control group - CG), 250UI (GI) or 500UI (GII) of BT-A under transabdominal ultrasound guidance. Semen was collected at baseline, 2, 4 and 8 weeks after treatment. Semen parameters were determined and seminal plasma pH, total protein (TP), total chlorides (TC), calcium (Ca), potassium (K), and sodium (Na) concentrations were assessed. One-dimensional sodium dodecyl sulfatepolyacrilamide gel eletrophoresis (SDS- PAGE) was performed to determine seminal plasma protein profile. Sperm parameters and seminal plasma pH, TP, TC, Ca and K mean values did not change significantly at any time point and among treated groups (P>0.05). The SDS-PAGE analysis of the pooled fractions identified 31 protein bands with molecular weights ranging from 3.9 to 106.2kDA in all treatment groups during the entire evaluation period. Regardless the used dose, intraprostatic BT-A injection do not alter semen parameters and seminal plasma biochemical and protein profiles of dogs with BPH.

A prostaglandina F2? pode ser usada em caes para aumentar o volume do ejaculado em casos de inseminação artificial, criopreservação seminal ou biotecnologias de reprodução. Os efeitos colaterais após a administração da PGF2a, como taquicardia, salivação, emese, diarréia e convulsões geralmente são relacionadas com a dose utilizada. Esse trabalho objetiva relatar a ocorrência de hepatite tóxica aguda após a administração de PGF2a em um cão, e discutir a importância de se utilizar essa droga com cautela nessa espécie.

Prostaglandin F2? can be used in dogs to increase ejaculate volume in cases of artificial insemination, semen cryopreservation or reproductive biotechnologies. Side effects after administration of PGF2? in dogs as tachycardia, tachypnea, salivation, vomiting, diarrhea and seizures are usually dose- dependent. This paper reports the occurrence of acute toxic hepatitis after the application of PGF2? in a dog, and discusses the importance of using this drug with caution in dogs.

O objetivo do trabalho foi avaliar a taxa de maturação nuclear in vitro de oócitos provenientes de gatas doméstica púbere e pré-púbere. Foram utilizadas 15 fêmeas felinas, 10 púberes e 5 pré-púberes; sendo os oócitos obtidos por aspiração quantificados e classificados. Os oócitos classificados como excelentes e regulares foram reunidos em grupos de 10, em meio de cultura, recobertos em óleo mineral em Placas de Petri siliconizadas e descartáveis. Após permanência em estufa, a 38°C e 5% de CO2 por 48 horas, os oócitos foram submetidos a duas lavagens com solução de hialuronidase a 0,4%, fixados em metanol/acido acético e corados com orceína acética. A avaliação da configuração cromossômica de oócitos maturados in vitro resultou em 44,68% das células em metáfase II no grupo das fêmeas púberes e 25,32% no grupo das doadoras pré-púberes, indicando que a puberdade influencia a capacidade dos oócitos se desenvolverem in vitro.

This study assessed the in vitro oocyte nuclear maturation in adult and young domestic cats. Fifteen ovaries were used; 10 from adult females and 5 from young females. The oocytes collected by aspiration were quantified and classified. The oocytes classified as excellent and/or regular were grouped (10 oocyte /drop) in culture medium covered with mineral oil in disposable Petri dishes. The oocyte were incubated at 38°C and 5% de CO2 for 48 hours and then washed with 0,4% hyaluronidase, fixed in methanol/acetic acid and stained with acetic orcein. Evaluation of chromosomal configuration of oocytes matured in vitro showed 44,68% of oocyte in metaphase II in the adult female group and 25,32% in the young donor group, showing that puberty influences oocyte capacity for in vitro development.

Com o objetivo de avaliar a função reprodutiva de 4 cães machos, adultos, a citologia aspirativa com agulha fina (FNAC) e as concentrações de testosterona sérica foram usadas em associação a determinação do volume testicular e análise do sêmen. FNAC foi realizada em ambos os testículos e as concentrações de testosterona foram determinadas durante 24 horas, em intervalos regulares. Os resultados da análise do semen se encontraram dentro do padrão para a espécie nos cães 1 e 3. O cão 2 apresentou testículos pequenos, baixa qualidade seminal, alta porcentagem de células de Sertoli e espermátides iniciais sugerindo uma degeneração testicular. No cão 4 observou-se uma degeneração testicular do lado direito conseqüência de um processo obstrutivo mostrada pela diminuição do testículo, baixa qualidade do sêmen evidenciada pela baixa concentração espermática e incontável número de espermatozóides na FNAC; uma diminuição do trânsito espermático foi observada no testículo esquerdo, com espessamento do epidídimo, alta porcentagem de gotas distais na análise seminal, porém resultados normais na FNAC. O ritmo circadiano da testosteronafoi claro nos cães 3 e 4, entretanto as concentrações encontraram-se próximas ao limite inferior. O volume testicular, a análise do sêmen e a FNAC testicular podem fornecer informações valiosas sobre a espermatogênese. Entretanto, as concentrações séricas de testosterona não são claramente correlacionadas as características reprodutivas nesses cães.

In order to verify alternative clinical approach in the reproductive evaluation of 4 adult dogs, the fine needle aspiration cytology (FNAC) and serum testosterone levels were used together with testicular volume measurement and semen analysis. FNAC was performed in both testes and serum testosterone concentrations were assayed in regular intervals during a 24h period. Results of semen analysis and FNAC were normal in dogs 1 and 3. In dog 2, small testes, poor semen quality, a high percentage of Sertoli cells and early spermatids were found suggesting testicular degeneration. In dog 4, a small right testicle, poor semen quality with low sperm concentration and an uncounted amount of spermatozoa in the FNAC indicated testicular degeneration due to an obstructive lesion; whereas high percentage of distal droplets, thicker left epididymis and normal FNAC of the left testis suggested a slow sperm transit. Testosterone circadian rhythm was clear in 3 of 4 dogs, although concentrations were low. Testicular volume, semen analysis and testicular FNAC could provide valuable information about spermatogenesis. In contrast, serum testosterone concentration was not clearly correlated with any reproductive characteristic of those dogs.

O presente estudo objetivou diagnosticar as causas da morte embrionária precoce em 128 éguas. Amostras de soro foram coletadas nos dias 4, 7, 10, 13, 16, 19, 21 e 30 após ovulação, para dosagens hormonais e mensurações do corpo lúteo. O diâmetro e as características da vesícula embrionária foram avaliados, através da ultra-sonografia, a partir do 12º dia pós-ovulação. Das 128 éguas estudadas, 17 (13.28%) apresentaram morte embrionária. O diâmetro do corpo lúteo, bem como as concentrações plasmáticas de progesterona, foi semelhante nos grupos que apresentaram morte embrionária e nos que mantiveram a gestação. Os níveis de estrogeno plasmático foram mais elevados no grupo das fêmeas que mantiveram a gestação. Os exames citológicos, microbiológicos e histopatológicos revelaram que a maioria das éguas com diagnóstico de morte embrionária eram portadoras de endometrites.

The present study aimed to diagnose early embryonic death in 128 mares. Blood samples were collected at the 4th, 7th, 10th, 13th, 16th, 19th, 21st and 30th day after ovulation and the diameter of the corpus luteum was measured. The diameter and characteristics of the embryonic vesicle were evaluated by ultrasonography. Microbiology, cytology and histopathology were carried out in mares with embryonic death. Plasma oestrogen and progesterone concentration were measured by radioimmunoassay. From 128 mares, 17 (13.28%) showed early embryonic death. The embryonic losses took place at 19 days of pregnancy in 47.05% and at 21 days of pregnancy in 29.4% of the mares. The diameter of the corpus luteum in the mares that maintained pregnancy was similar to those with embryonic loss. Otherwise, the diameter of the embryonic vesicles was bigger at the 16th, 19th, 21st and 30th day of pregnancy when compared to the mares with embryonic loss. The mean plasma progesterone concentration was similar in both groups and the median plasma oestrogen concentration was higher in the pregnant mares. The cytological, microbiological and histopathological exams revealed that most of the mares had endometritis. Ultrasonography provided an early diagnosis of pregnancy (from the 12th day post-ovulation) and important information about the development of embryo and embryonic death. Endometritis was considered the main cause of embryonic losses in this study.
